La placenta humana es un órgano altamente especializado, esencial para el desarrollo fetal por sus funciones endocrinas, metabólicas e inmunológicas. Su estudio en modelos animales presenta limitaciones debido a diferencias estructurales y funcionales entre especies. Los cultivos de explantes placentarios humanos representan una herramienta experimental valiosa, ya que preservan la arquitectura tisular y las interacciones celulares, permitiendo estudiar mecanismos fisiológicos, fisiopatológicos y farmacológicos bajo condiciones controladas. Objetivo: Desarrollar, optimizar y caracterizar un modelo de cultivo de explantes placentarios humanos del tercer trimestre mediante la evaluación integrada de la viabilidad, diferenciación e integridad morfológica e inmunológica. Métodos: Se recolectaron cinco placentas de mujeres nulíparas sanas, obtenidas por cesárea o parto vaginal. Los explantes fueron cultivados durante 7 días en medio RPMI-1640 suplementado a 37 °C, 5% CO₂ y 10% O₂. La viabilidad se evaluó mediante la actividad del lactato deshidrogenasa (LDH), la diferenciación mediante la secreción de la fracción beta de gonadotrofina coriónica humana (β-hCG) y la integridad tisular mediante inmunohistoquímica para: β-hCG, lactógeno placentario humano (hPL), fosfatasa alcalina placentaria (PLAP), CD45⁺, CD68⁺ y CD3⁻. Resultados: La actividad de LDH mostró un aumento al día 2, disminución hasta el día 5, incrementando al día 7. La β-hCG alcanzó su máximo al día 4, sugiriendo diferenciación trofoblástica activa. La inmunohistoquímica confirmó la expresión de β-hCG, hPL y PLAP. Conclusiones: El modelo demuestra viabilidad reproducible y estabilidad funcional durante 7 días. Constituye una plataforma útil para estudios de inflamación, estrés oxidativo, hipoxia y evaluación farmacológica en tejido placentario humano.
