Resumen : La marchitez bacteriana, causada por Ralstonia solanacearum (Smith) Yabuuchi et al, es una de las enfermedades mas importantes en el cultivo de tomate (Solanum lycopersicum L.). En los últimos años se ha venido estudiando el uso de extractos naturales y microorganismos antagonistas en el combate de la enfermedad; sin embargo, las investigaciones se han centrado en los mecanismos de acción de extractos vegetales y de microorganismos antagonistas sobre la bacteria patógena; pero hay poca información acerca del comportamiento poblacional de R. solanacearum en el suelo y en la planta, en presencia de extractos vegetales y de microorganismos antagonistas o en ausencia de estos. Los estudios se concentran en la manifestación de la marchitez bacteriana en las plantas de tomate, sin tomar en cuenta el efecto que producen los extractos vegetales y/o las bacterias antagonistas sobre las poblaciones en el suelo de R. solanacearum y la relación que hay entre este efecto y la manifestación de la marchitez bacteriana. ¿Si se conoce la dinámica poblacional de la bacteria en el suelo, se podrá predecir el ataque al cultivo? Cuantificar y relacionar matemáticamente, permitiría introducir elementos de juicio que pudieran ayudar a aclarar el mecanismo de acción y/o las causas por las cuales hay respuestas diferenciales al uso de extractos vegetales y bacterias antagónicas. En el presente estudio se trabajará en el modelado matemático de poblaciones de R. solanacearum, y de la manifestación de la marchitez bacteriana del tomate, tanto en ausencia como en presencia de bacterias antagonistas y/o extractos vegetales. Relacionando la dinámica poblacional en el suelo, la manifestación de la enfermedad en la planta y la respuesta a los diferentes tratamientos considerados en el estudio, con la intención de hallar indicios que permitan inferir sobre los mecanismos de acción o disminuir la variabilidad de las respuestas, dado el establecimiento de los modelos de predicción matemáticos. La investigación se desarrollará en el Laboratorio de Bacterias Fitopatógenas de la Facultad de Agronomía de la UCV Campus Maracay. Se diseñará un experimento para obtener datos del comportamiento de R. solanacearum en el suelo y de la manifestación externa de la Marchitez Bacteriana en plantas de tomate en diferentes condiciones. Se usarán aislamientos de R. solanacearum, P. fluorescens y B. subtilis provenientes de la colección del Laboratorio de Bacterias Fitopatógenas, extractos acuosos de especies vegetales comunes en el Campus Maracay de la UCV: Heliotropium indicum L. (Rabo de Alacrán) y Ricinus communis L. (Tártago) y el producto comercial Timorex Gold, basado en el extracto de la planta Melaleuca alternifolia. El experimento se llevará a cabo en un diseño completamente aleatorizado, con 7 tratamientos y 3 réplicas. Los tratamientos corresponden a las bacterias antagonistas, los extractos vegetales y el producto comercial que se aplicarán a las plantas, bajo cuya influencia será medido el comportamiento de la bacteria en el suelo y de la enfermedad en el cultivo; además de dos testigos. La inoculación se realizará al momento del transplante y, a partir de este, cada 5 días se cuantificará la población de R. solanacearum en el suelo de cada maceta, el grado de severidad de la marchitez bacteriana en cada planta de tomate y el número de plantas enfermas en cada tratamiento. Los datos de UFC/g de suelo, de IE y de IIM, se graficarán y se enunciarán los modelos matemáticos que mejor describan la dinámica poblacional, en el suelo, de R. solanacearum y de la marchitez bacteriana, en las diferentes condiciones estudiadas. Adicionalmente, se procederá a correlacionar la dinámica poblacional, en el suelo, de R. solanacearum (UFC/g de suelo) con la manifestación de la marchitez bacteriana en las plantas (IE e IIM).
Descripción : Bacterial wilt caused by Ralstonia solanacearum (Smith) Yabuuchi et al, is one of the most important diseases in the cultivation of tomato (Solanum lycopersicum L.). During the last years has been studying the use of natural extracts and antagonistic microorganisms in combating the disease; however, research has focused on the mechanisms of action of plant extracts and antagonistic microorganisms on pathogenic bacteria; but there is little information about the population behavior of R. solanacearum in soil and in the plant, in the presence of plant extracts and antagonistic microorganisms or in their absence. The studies focus on the demonstration of bacterial wilt in tomato plants, regardless of the effect produced by the plant extracts and/or antagonistic bacteria on populations in the soil of R. solanacearum and the relationship between this effect and the demonstration of bacterial wilt. If the population dynamics of the bacteria in the soil is known, is able to predict the attack on the crop? Quantify and relate mathematically, would introduce evidence that could help clarify the mechanism of action and/or the reasons why there are differential responses to the use of plant extracts and antagonistic bacteria. In the present study we will work on the mathematical modeling of populations of R. solanacearum, and the demonstration of bacterial wilt of tomato, both in the absence and presence of antagonistic bacteria and/or plant extracts. Linking population dynamics on the ground, the manifestation of the plant disease and response to different treatments considered in the study, with the intention of finding evidence to support the mechanisms of action or reduce the variability of the responses, since the establishment of mathematical prediction models. The research was conducted in the Laboratory of plant pathogenic bacteria of the Faculty of Agronomy of the UCV Campus Maracay. R. solanacearum, P. fluorescens and B. subtilis isolates will be used from the collection of the Laboratory of plant pathogenic bacteria, aqueous extracts of common plant species in the UCV Campus Maracay: Heliotropium indicum L. (Scorpion's tail) and Ricinus communis L. (Spurge) and Timorex Gold commercial product based on the extract of the plant Melaleuca alternifolia. The experiment was conducted in a completely randomized design with 7 treatments and 3 replicates. Treatments correspond to antagonistic bacteria, plant extracts and commercial product to be applied to plants, under whose influence will be measured behavior of the bacteria in the soil and crop disease; and two witnesses. Inoculation was performed at the time of transplantation and, from this, every 5 days the population of R. solanacearum be quantified in the soil of each pot, the severity of bacterial wilt in each tomato plant and the number of diseased plants in each treatment. Data from CFU/g of soil, IE and IIM, will graph and mathematical models that describe the population dynamics in the soil, will be stated R. solanacearum and bacterial wilt in the studied conditions. In addition, shall be correlated population dynamics in the soil, R. solanacearum (CFU/g of soil) with the manifestation of bacterial wilt in (IE and IIM).
